DNA-Free HS LATaq DNA polymerase is a HotStart LATaq DNA polymerase, which requires activation by heating at 95℃ for 5 minutes or 98℃ for 2 minutes. LATaq DNA polymerase is a genetically engineered enzyme based on Taq polymerase, containing a double-stranded DNA-binding protein segment that significantly enhances the polymerase's extension capability and speed. Therefore, LATaq DNA polymerase is highly suitable for amplifying long DNA fragments, with fragment lengths reaching 5-8 kb and an extension speed of 3-4 kb/min (at 72℃). This enzyme is a recombinant DNA polymerase expressed in Escherichia coli, with a molecular weight of 107 kDa. It exhibits 5’→3’ polymerase activity, weak 5’→3’ exonuclease activity, and no 3’→5’ exonuclease activity. The amplification products have 3'-dA overhangs.

Our DNA-Free HS LATaq DNA polymerase, on the other hand, is devoid of genomic and plasmid DNA from the production strain. This makes DNA-free Pfu particularly suitable for amplifying, sequencing, and cloning 16S and 23S rDNA genes, as well as for standard applications like PCR and primer extension.

 

Unit Definition

The amount of enzyme required to incorporate 10 nmol of deoxyribonucleotide into acid-insoluble material within 30 minutes at 72℃ using salmon sperm DNA as the template/primer is defined as one unit of activity (U).
